📖 ABSTRACT/OVERVIEW
The development of a comprehensive Nigerian Herbal Pharmacopoeia depends on the availability of rigorous standardisation data for locally significant medicinal plant species. Daniellia oliveri, a tree resin and bark-providing species distributed across North Central and North West Nigeria, lacks formal pharmacopoeial monograph data despite widespread traditional use. This study establishes comprehensive pharmacognostic standardisation parameters for Daniellia oliveri bark and gum resin drug materials to support their inclusion in the Nigerian Herbal Pharmacopoeia. Samples were collected from Kwara, Niger, and Kogi States, representing different ecological sub-zones within the Guinea savannah belt. Standardisation parameters evaluated included macroscopic and microscopic description, moisture content, total ash, acid-insoluble ash, water-soluble extractive, ethanol-soluble extractive, volatile oil content, and foaming and swelling indices in accordance with WHO monograph guidelines. HPTLC fingerprinting was conducted as an identity verification tool. Phytochemical screening confirmed the presence of terpenoids, phenolics, resins, and flavonoids. Inter-source variation in extractive values and volatile oil content was observed and documented. Microscopic analysis revealed distinct diagnostic features including resin canals and calcium oxalate crystals. The standardisation data package compiled in this study constitutes a complete pharmacognostic monograph for submission to the Nigerian Natural Medicine Development Agency for pharmacopoeial consideration.
