Evolutionary Dynamics of Pyrethroid Resistance in Anopheles gambiae s.l. Across an Insecticide Use Gradient in South West Nigeria: A Genomic and Phenotypic Investigation

The rapid global spread of pyrethroid resistance in Anopheles gambiae s.l. threatens the efficacy of insecticide-treated nets and indoor residual spraying, which remain the primary tools of the Nigeria National Malaria Elimination Programme. Understanding the evolutionary trajectory of resistance requires both phenotypic characterization and genomic analysis of selection signatures, yet such integrated studies are absent for South West Nigeria. This study investigates the evolutionary dynamics of pyrethroid resistance across a spatial insecticide use gradient spanning urban Lagos, peri-urban Ogun, and rural Ondo states in South West Nigeria. WHO cylinder bioassays and concentration-response assays will quantify resistance intensity at 12 sites. Whole-genome sequencing of 240 An. gambiae s.l. specimens will enable genome-wide selection scans using XP-EHH and FST statistics to identify genomic regions under positive selection for resistance. The contribution of kdr L1014F, L1014S, and N1575Y alleles alongside CYP metabolic gene amplifications will be assessed. Coalescent modeling will estimate the age and geographic origin of resistance alleles in the study area. This study makes an original contribution by applying population genomics to characterize selection dynamics of resistance in a major Nigerian malaria zone, directly informing resistance management planning.
